Having just started running after having done a few Sportives/Audaxes, I'd suggest the following advice:

Don't forget the benefit of riding in a group, sharing the work at the front. There is a tremendous benefit to be gained from drafting someone. But if you can go faster on your own than tucked behind a slower rider, go on ahead.

There will be stops for drinks/food. Take advantage of these, but don't stop too long. Fill up water bottles, and have some food - flapjacks, cake, malt loaf and bananas are all recommended.

Take some food with you. I always go for Jelly Babies - cheaper and less messy than energy gels.

Make sure you bike is in good working order, tyres pumped up, chain oiled, and spare inner tube or two on board before you start.

Start slowly. On my first Sportive it felt more like an afternoon meander than a challenge for the first mile. Pick up you speed after that, but leave some in the tank for the end.
