On my caad8 with 105 5700 10 spd groupset, there is just one gear change , 50 teeth front chainring with 3rd cog down from the 34 cog on the cassette, if I change down to it from the 4th cog on cassette, it goes click click click click until it finally makes the change. Like I said this the only gearshift where this happens, the rest work perfectly. Is this a twist of the barrel adjuster on rear mech to cure and if so, which direction do you turn it ?

My thoughts are, if all the other changes are working ok, if you alter the cable tension, then it could upset all the other changes.

One possibility is that the inner gear cable is sticking slightly at that point. Replacement of the inner and outer gear cable might be the answer?

To loosen the tension you turn the adjuster clockwise.
But first, try undoing the inner cable clamp at the RD and see if the cable is moving smoothly within the outer. It may be as @Sharky says new inner and outer cable time

what he said, but in my own words:
screw adjuster in - the cable gets less tension: Spring pulls the derailleur towards the small cogs
Screw it out - more tension, pulling the derailleur towards the large cogs.
