I haven't tried it but IMO it is not something I would recommend. It will likely flap around something awful and possibly increase the chances of pulling the bikes off the rack. You are effectively turning the bikes into a large sail!
Not sure I'd do it with a car for the reasons skol states. Even with the air pocket created by a motorhome if you forget to bungee it around the bikes it could have catastrophic consequences on a motorway, guess how I know this. Using the reversing camera as a rear view mirror saved the bikes but I still had to sacrifice a stepladder to make plates to repair the rack fixing points
Never bothered covering my bike when its on the rack just remove all the water bottles, Gps computer, pump and the like, i also tie the mud guards to the wheel to prevent them flapping about.
I will be using the bike rack on the car and camper.
I'm not bothered about the bike getting rain wet its the extra crud thrown up by the rear wheels etc.
