I think I have finally solved the sore ass issue! 
The current saddle is a ladies Specialized Sonoma gel saddle. It was rubbing a bit at first and I thought it would be getting swapped out for something else.
I re-adjusted the height, tilted it by 1mm and it was a lot better on the last ride, although the inside of one of my thighs felt a little sore.
Before I rode today I checked everything again and the seat was off centre by a few mm.
I set it straight, in line with with the handlebars, and that has done the trick.
Combined with padded shorts that now fit me (others were too big) I now finally feel comfortable!
