I did it this summer on a tourer. What ever bike you use I would suggest getting a bike fit, thats what I did and I had no aches or pains from positioning at all. I would also suggest researching the ride but not too much. At the end of the day everyones is different and it is as good as you make it. Don't be afraid to change your route as you are doing it either.
Take your time and enjoy it, it is bloody hard but very rewarding and great fun.
