I can't help you with the full route, but last year I did from Chertsey, on the edge of London down to Portsmouth. The off-road section using NCN221 along side the Basingstoke Canal was particularly pleasant I remember, and had I been heading for Southampton, I'd probably have taken it further.
Depends on the kind of cycling you prefer doing, but there are a couple of NCN options you could pick up from Southampton.
