I've been cycling on and off for a few years, my longest is 55 miles, and I'm currently on a GT Traffic 1.0 hybrid. It's quite quick and fitted with Ergon grips to help out with hand positions.

I've recently just started again and doing 20-25 miles. My brother (also just started) has suggested this sportive in September. There are routes of 47, 68 and 100.

In 3 or 4 months, could I get up to the required fitness for 100 miles, or should I play it safer at the 68, or even 47?

Sign up for the 100. It will give you a target to aim for. If you up your mileage in chunks of 20 miles, over the next months, you should be able to do it, no problem. If you find you really can't manage it on the day, just detour onto a shorter option.

A jump up to 100 miles from 20-25 in 3-4 months is too big a jump, unless you can get out and train every day. But as 55 miles is your furthest, and shining you can only get out at weekends and occasional evenings, I'd suggest a target of achieving 68 wouldn't be unrealistic, if you increase your distances by around 5 miles or so every time you go out.

If you can ride 25 miles, you can ride 100 miles. The thing you need to ask yourself is do you want to ride it fast or at a Plod. 100 miles is a long way.

I would be heading for a fast 47 miler. 3 or 4 months training would bring you up to that. Increasing your training in 20 mile chunks is a massive ask, in my opinion. 5 mile increases is achievable and you can also add speed and climbing into that. At least you will finish the day with a decent ride and not struggling to finish.
