For those that don't know this goes from Chepstow to Fishguard across South Wales along mainly sustrans route 4 and 47. Has anyone cycled it on a road bike? There are a few sections described as traffic free, so I'm wondering if the surface of these is suitable for a road bike.

I've cycled some but not all it, so can't comment on every section. Traffic free sections of NCN routes can vary from smooth and well tarmacked, to rocky and rutted - and sometimes cunningly change from the former to the latter after you've gone a few miles and it is too late to turn back.

Having said that my memory of the sections in South Wales were that they were OK, and in general I've rarely come across a traffic free NCN route that I can't do on my tourer with 28mm tyres.
