I've got a wee sneaky desire for this bad boy, mostly because my wife has the little brother version and I've always cast a bit of an envious eye over it. A short stem and compact bars would reduce the strain on a week back, and a measure from a fitter to make sure your cleats are in line would reduce the strain on the knees as well as the more obvious saddle height selection.

I really like my Genesis Equilibrium brought from on here.
If I could only have one bike though, this wouldn't it (would be second choice). My first choice is my Raleigh Revenio ☺
It's really comfortable to ride all day and riding on the flat bar I'm almost sat upright

The Giant Defy is very good for a less flexible rider, my wife rides the men's Defy 1; She's tall and of a heavier build and not especially fit at the moment, but she got it to build up cycling fitness and she's finding the bike ideal. It's comfy as anything geometry wise, and still a very good road bike.

Di2? That'll do nothing for your comfort or bike fitness... I think that's established, but I thought I'd add to the weight of opinion. :-)

get an aluminium bike:

Allez DSW SL Sprint Expert

The Allez DSW SL Sprint Expert is one of the most advanced alloy road bikes Specialized have ever made, with the focus of being a reliable crit-racing weapon. At the heart of the DSW Sprint Specialized have added more or taken away material where necessary, resulting in an aluminium bike that's more compliant and considerably stiffer than ever before. How stiff, exactly? It's the stiffest alloy bike Specialized have ever tested. Drawing inspiration from the Rider-First Engineered Tarmac, this frame has unique aerodynamic tube shapes, bottom bracket shell, head tube and fork. This ensures uniform performance across the entire size run meaning every single watt you produce gets transferred to the road.

I'm looking for a good used one

That looks a really aggressive race position @dim
Not sure it's one for all day riding, but I personally like the look of it

a longer stem for the handlebar, and it will become more relaxed. The pic that I posted shows a big drop in the saddle height and handlebar .... I'm looking at a medium frame (52-53cm), and I doubt that the saddle to handlebar height will be as high as in the pic

The OP has a very good bike though and I'm not sure that the Allez sprint expert will be better, but I thought that I'd throw in a good aluminium bike as a suggestion to others.

Perhaps it will be a lot more robust than a carbon framed bike, and it weighs as much

Go up to Ribble and try a 525 mine is lovely and a nice relaxed geometry. I would recommend you try to borrow something with a more audax/ sportive feel and try doing 10 miles see if it is less hurty.
