Am going to Paris for one night and two days next week.

Will be staying at a hotel near the Gare de l'est and was planning on mainly walking around.
Arrive at 9am on Eurostar then leave the next day at 8pm.

Any ideas for what I should do, first time there?!?!?

And how does that bike hire thing work?

Musee d'orsay should be the first objective for your daytime activity.

If you're near the Gare de l'est then there are some trendy / earthy bars around the the 10th arrondissment. I'd honestly just ask some trendy looking people walking around the 10th where a good bar was. It's a bit cliquey around there though - a bit like a newly yuppified london borough.

For a definite good drink / kebab, albeit a bit touristy, then the latin quarter.
 
The Grande Arche at La Defense is always good value - great view of the city from the roof and interesting galleries inside etc...
Tour Montparnasse - see Paris from 55 floors up...
Parc Andre Citroen is worth a visit, as is Sacre Coeur and the Place du Tetre, walking North and up for more spectacular views.
And the usual monuments of course. Try a Chocolat a l'Africain at Angelinas opposite the Louvre - Rue de Rivoli. Powerful stuff!
Velib bikes are 20 odd kg and made by Mercier! You need a credit card to subscribe and use - never done it though.

Galeries Lafayette has great (free) views from the roof (and a nice brasserie up there too). And then you can pretty much walk in any direction from there and soak up the city.

I haven't been to Paris in ages - I really must go back soon.

If you like something a bit different - try the Paris Catacombs ... tube station is Denfert Rocherau.
