I prefer the lane route myself (NCN 41 to the bridge, then St Briavels/Bream/Parkend/Soudley/Littledean/Northwood Green/Tibberton/Highnam), but neither the A38 nor the A48 are bad, though it may be best to avoid the Gloucester area during rush hour.
Approaching Gloucester on the A38, take the first exit off the dual and go through Quedgeley to the lights at the other end of the dual. There's roadside cycle track for all of the SW bypass and A40 from these lights to just after the A40/A48 roundabout. The stretch from the Hempsted roundabout to just before the lights by the docks is unreconstructed footway - a bit bumpy and prone to get cars parked on it, but the rest is good.
