After a mechanical issue a month back, I replaced my wheels (Fulcrum 5), dérailleur (105), hanger and chain. I am about 200 miles into this new setup.

Every now and then when in a high gear / riding fast, or if I pull away in too high a gear, I'll hear a click or clunking noise from the rear of the bike, very short, like something is tightening (similar noise to what the cassette lockring made when I tightened it).

Everything seems tight, and the cassette is in good condition. Is this perhaps just spoke tension from the new wheels or is something wrong?

Try putting a little lube/grease in between the spokes of the rear wheel where they cross and see if that solves the problem.
If not Bottom bracket?
The noise may seem to be coming from the rear but I had a similar problem and discovered to my surprise that the bottom bracket was on its way out.
