The + figure is height gained (climbing) and the - figure is height lost (descending), so total climbing = 22520 ft. BTW, if you plot the route from scratch in ridewithgps, it will add turn by turn instructions that you will see in a pane down the left, these will display on the Garmin in a shorthand format to give clues on where to go satnav style.
IME, a typical flat 100 mile ride usually involves between 3 and 4 thousand ft elevation gain, all the small undulations add up, even if there are no proper climbs, so for your 300+ mile route you are looking at 9-12 thousand ft assuming it is flat, if it is not all flat, i.e. there will be some hilly sections you pass through (say a 50 mile section or 2, with 4 or 5 thousand ft climbing), the figure stated by ridewithgps doesn't look otherworldly IMO.
12,000ft over 300 miles is what I would call pancake flat, chances are in the UK you won't go 300 mile without hitting some hilly sections.
