I had a tyre look like that after a nasty pothole smack. It didn't damage the rim but the tyre was, well, I think the phrase is FUBAR with that permowobble.. fit only for the bin.

Tyre replaced. Now I can't identify the spot on the tyre?

....
I don't see how that would be possible.

cheap tube pushing the tyre away from the rim at the valve possibly

Tyres are composites. At the most basic, they are a rubber-threads-rubber sandwich. The threads give strength and stop the tyre expanding in the same way as the tube that fills it. The rubber provides a wearing, gripping face, and holds the threads in alignment so that they can do their job.

It was more a case of the inner tube being very constricted and much bigger at different points -made the tyre sit unevenly and was quite noticeable. Took me a time to find out what the problem was too, it was a bit of a puzzler. It was the only inner tube I've ever had a problem with as well (thankfully).

I don't see how that would be possible.
