Sore backside after 50 odd miles

I thought I had my backside comfort sorted: Santini twistgel shorts, selle italia turbomatic saddle, assos cream.

The same as I've been using for a while, although these shorts are only a few rides old. Did 70 miles a few weeks ago no bother, plenty of rides of up to 50/60 miles but today when pushing to do 80 miles I got a seriously sore nether regions at about 60 miles. The last 20 were agony, lifting myself off the perch at every opportunity :ohmy: it was not at all pleasant. My concern is "how the hell am I going to complete the ride London 100?"

Any suggestions?
 
Basically, it's about your sit bones adjusting to the saddle, regardless of what shorts you have.

Is it a new bike? If so, do you still have your old bike/saddle? Otherwise, you'll just have to grit your teeth and get the miles in your legs (and pain in your backside) but you should be able to break it in. If it just remains a real pain in the ar*e, you might have to try and find an alternative saddle.

Thanks for the reply,and the link. The saddle is the same make and model I had on my old bike before it was written off in an accident, the original saddle rails broke in the impact. I've been using this one for about 4-5 months or so and all was fine until I upped the mileage, it was OK up to 60 -70 miles, but today, jeez no way! The bike is a boardman team carbon and I very quickly realised the fizik arione wasn't for me, so I sought out a selle italia turbomatic, identical to the one I had before, except this one wasn't the team edition. I can't work out why it became so painful though.

I am considering getting a saddle fit using the selle italia ID match system. Pedal Heaven, a shop local to me in Fleet, offer this and the specialised option, I'd like to know if my saddle actually is right for my backside.
