I have a question to ask.

I have microsoft office and on e-mail I have blocked someones e-mail address as they are a pain in the arse. If they e-mail me again will they get a message back saying they have been blocked or mail undeliverable or will they just be under the assumption that the mail has been sent and none the wiser????

Don't know, what usually happens with your e-mails?;)
 
If you just add it to the blocked senders list, it just dumps it in your junk mail box but doesn't send a reply. You can create a rule which will send a reply if you want (just right click on the e-mail and create rule).
 
If your ISP offers spam filtering then you can add the sender to a blocked list via the webmail page and it will never reach you. If you wish them to know you are blocking them a single email should suffice so I assume you don't want them to know this.

If you have blocked a senders email address in your mail application the most likely out come is that the email will be deleted or moved to trash.
The sender receives nothing so as far as they know you are reading messages sent.
