My rear derailleur was being slow at changing down so I stupidly decided to "adjust" it.... Now it's far worse than when I started.

So the current situation is the rear barrel adjuster doesn't turn so I was using the one on the handlebars. If I start in the highest rear gear it doesn't change down for the first two or three clicks but then almost makes it to the lowest gear. Does that mean that the cable needs to be tighter at the pinch bolt?

Sounds more like the little bit of outer down by the mech needs changing, probably needs a new inner too.

If by 'highest' you mean smallest sprocket, then yes, failure to shift probably means the cable is too slack.
